Factors to consider when selecting prefilled syringe components
Selecting components for a prefilled syringe requires an assessment of the entire drug–container–device system. Pharmaceutical teams should consider the following factors before specifying a solution.
Drug formulation and compatibility
The characteristics of the drug formulation influence which elastomeric compounds and coating options may be suitable. Sensitive biologics and complex injectable formulations may require components designed to minimize extractables and leachables, reduce particulate risk and limit interaction between the drug and elastomer.
Compatibility should be supported by appropriate testing rather than assumed from the component material alone.
Syringe format and device compatibility
Plungers and front-end closures must be matched to the syringe barrel and intended delivery device. Relevant considerations can include component dimensions, container closure integrity, break-loose force, glide force and performance after storage.
For syringes used in autoinjectors or other device-assisted systems, components should also be evaluated within the complete device configuration.
Sterilization and supply format
Sterilization compatibility varies by component, compound and product configuration. Steam, gamma and ethylene oxide sterilization options are available across Datwyler’s component range where applicable.
Teams should confirm that the selected component and supply format—such as ready-for-sterilization, ready-to-use or Rapid Transfer Port packaging—are compatible with the intended fill-finish process.
Development stage and scalability
Component requirements can change as a program moves from early development and clinical supply to commercial production. Teams should consider the availability of technical data, packaging formats, production capacity and supply continuity from the beginning of development.
Selecting components with a viable path to commercial scale can help reduce the risk of late-stage requalification.
Datwyler’s prefilled syringe component portfolio
Datwyler’s portfolio includes plungers, needle shields and tip caps developed to support drug compatibility, container closure integrity and reliable syringe performance.
Plungers
Datwyler offers plungers for standard prefilled syringe formats from 0.5 mL to 10 mL, with custom sizes available for selected applications.
Options include FM257 and FM457 elastomeric compounds and NeoFlex™ fluoropolymer-coated plungers for sensitive formulations. NeoFlex™ creates a barrier between the elastomer and drug product, helping minimize extractables and leachables, reduce sub-visible particle levels and provide silicone-free plunger performance.
Plunger selection should account for syringe dimensions, formulation compatibility, container closure integrity, break-loose and glide performance, and integration with any secondary delivery device.
Needle shields
Soft and rigid needle shields help protect pre-attached needles and maintain the sterile barrier during manufacturing, filling, transportation, storage and handling.
Datwyler offers needle shield options in different formats and compounds, including FM30. The appropriate solution should be selected according to the syringe configuration, processing requirements and applicable sterilization method.
Tip caps
Tip caps provide front-end closure for luer-tip prefilled syringes. Datwyler offers ribbed and mushroom designs in selected compounds and configurations.
These components are designed to support sterile barrier protection, processing and handling. Sterilization compatibility should be confirmed for the selected design, compound and supply format.
Coated components for sensitive formulations
NeoFlex™ plungers use a full-surface fluoropolymer spray coating to create a protective barrier between the elastomer and drug product.
This technology is intended to minimize drug–component interactions, reduce extractables and leachables, lower sub-visible particle levels and support consistent functional performance. Coated components may be particularly relevant for biologics and other sensitive injectable formulations.
Manufacturing standards for prefilled syringe components
Datwyler’s prefilled syringe components are available under Advanced and FirstLine® manufacturing standards. The appropriate standard depends on the product’s quality, cleanliness, processing and supply requirements.
Advanced manufacturing
The Advanced standard supports pharmaceutical production through controlled manufacturing processes, automated production technologies, inspection capabilities and validated cleaning processes.
Available products and configurations should be confirmed according to the specific component and program requirements.
FirstLine® manufacturing
FirstLine® is Datwyler’s most advanced manufacturing standard. It combines fully integrated cleanroom environments, automated production, validated washing processes and camera inspection within a zero-defect manufacturing philosophy.
FirstLine® is designed for applications requiring particularly high levels of cleanliness and contamination control, including sensitive biologics and complex injectable formulations.
Datwyler’s technical team can help determine which manufacturing standard and product configuration are appropriate for a program’s requirements.
Support throughout prefilled syringe development
Datwyler combines component manufacturing with material expertise, technical support and a global production network.
Global manufacturing and supply support
Datwyler operates manufacturing facilities across Europe, North America and Asia. This footprint supports regional supply options, business continuity planning and commercial scalability.
Specific production locations and dual-sourcing options should be confirmed for the selected component and manufacturing standard.
Material and component expertise
Datwyler develops proprietary elastomeric compounds and coating technologies for injectable drug packaging. Its specialists can support component selection based on formulation sensitivity, syringe format, device requirements and processing conditions.
Quality-focused manufacturing
Datwyler’s manufacturing standards incorporate automation, inspection and controlled production processes. FirstLine® adds a fully integrated cleanroom environment for programs requiring particularly stringent contamination control.
These capabilities are supported by Datwyler’s zero-defect philosophy.
Technical and regulatory support
Datwyler provides technical documentation and data packages to support drug development and regulatory submissions.
This support can help pharmaceutical teams evaluate material, compatibility and component-performance considerations. Responsibility for qualification and regulatory approval remains with the drug and device manufacturer.
